标题: 3D Polymer-Graphene Biomaterials for Bone Tissue Regeneration
摘要:
Several tissue engineering strategies attempt to overcome limited self-regenerating capacity of human skeleton. The most common mimics the natural process of bone repair by delivering a source of progenitor cells and three dimensional biocompatible, bioresorbable and osteoconductive scaffolding matrices.
In this context we aimed at enhanced understanding and knowledge-based design of novel three-dimensional scaffolds based on polymer-graphene which provide an improved effectiveness and feasible alternative for current treatment options in the restoration of non-healing bone defects.
Molecular modelling tools were used to reduce the amount of poor design cycle paths. Subsequently, the materials and compositions suggested as most promising by means of molecular modelling were used for actual synthesis.
Using as a doorstep the molecular modeling results, porous CHT / graphene oxide (GO) scaffolds with 0.5, 1, 2, and 3 weight % GO were fabricated by freeze drying. The obtained biocomposites were extensively characterized regarding all important aspects to be used as scaffold for bone tissue engineering i.e. structural, morphological, degradation and ability to form hydroxapatite. The GO presence has a strong effect on the structural and morphological features of the biocomposites generating.
Advantages of adding GO in the structure of the scaffolds are revealed also in relation to cell viability, proliferation and attachment while histopathological analysis of the in vivo samples revealed larger amount of new bone in the CHT/GO 3.0 wt.%-filled compared with CHT and empty defect.

简介:
Dr. Mariana IONITA was educated at top level universities (Politecnico di Milano, Technical University of Denmark, Universita degli Studi di Milano or University of Pisa) and gained her PhD in Chemistry in 2008 and in Bioengineering in 2008. Currently she is carrying out very active research and teaching activities at Faculty of Medical Engineering contributing to advancement of knowledge in the graphene based biomaterials filed both computational and experiments, have been responsible for day-to-day co-ordination, consolidate the project planning, progress reports, or budgetary overviews, etc., for 8 research national / international projects. She conducted the assembly of an young but strong research team (5PhDs, 3postdocs, and 1 senior researcher), developed important  international collaboration, organized different international events and have been invited to and chair different international conference (e.g. ICCE-2015, IUMRS-ICAM2017, RICCCE-2017), appointed conference chair or board member of  conference (e.g. IUMRS-ICAM2017-Carbon Related Materials special Symposium and ICCE-NANO). She board member of Composite Part B Engineering and Nanotechnology Reviews scientific journals.
Her professional and scientific activity comprises research in the fields of biomaterials design for bone tissue engineering, with special focus on the development of complex protocols coupling molecular modeling and actual design to acquire improved solutions for bone regenerative medicine.
During the last years her research was focused on development of new optical / electrochemical graphene biosensor for testing osteogenic potency that provides a prompt ex vivo quantitative measure of the biological activity of mesenchymal stem cells with strong bioequivalence to bone formation in vivo.
